abstractsObjective To investigate the differential expression of proteins in extracellular exosomes of prion-infected cell models during prion clearance.Methods The proteins in extracellular exosomes of prion infected cell line SMB-S15 and its normal control cell line SMB-PS and SMB-S15 cells exposed to resveratrol for 4 days (RES4) and 8 days (RES8) were extracted and analyzed by isobaric tags for relative and absolute quantitation (iTRAQ) approach.Results A total of 4 809 proteins were detected from the extracellular exosomes of four cell lines,and their expression levels varied along with the clearance of prions,mainly down-regulated,which may be involved in important pathways including Parkinson's disease and Alzheimer's disease and Huntington's disease.Conclusions The proteins in the exosome changes in the process of clearing prions with resveratrol and pentose sulfate,suggesting that exosomes may be involved in the transmission of prions between cells.
